The $1.6 billion Toowoomba Second Range Crossing was officially opened to traffic on Sunday.
Deputy Prime Minister Michael McCormack said the 41km bypass would make transportation more efficient and take trucks out of central Toowoomba.
The Federal Government contributed $1.137 billion towards the cost of the project.
Premier Annastacia Palaszczuk said more than 3700 jobs had been created during construction.
She said the crossing would now play a vital role supporting ongoing growth of the region and State’s industries.
The Toowoomba Second Range Crossing links the Warrego Highway at Helidon Spa in the east to the Gore Highway at Athol in the west, via Charlton.
Construction included 10 million cubic metres of earthworks, 24 bridges and 40 road embankments.
